چکیده

The last sentence of the ‘‘WNV DNA Vaccine, Control Plasmid and E-protein’’ section of the Materials and Methods should read: ‘‘For protein booster vaccine formulation the E-protein was adjuvanted with 10 mg Matrix-M1 adjuvant (Novavax AB, Uppsala, Sweden).’’ In addition, the following sentence should be inserted as the last sentence of the ‘‘Vaccination Procedures’’ section of the Materials and Methods: ‘‘Mice were boosted four weeks later either via an IM injection of the same does of WNV-DermaVir nanoparticles or via subcutaneous (s.c.) injection of recombinant E-protein ([28]) formulated with Matrix-M1 (a kind gift from Novavax AB, Uppsala, Sweden).’’

Vaccination of Mice Using the West Nile Virus E-Protein in a DNA Prime-Protein Boost Strategy Stimulates Cell-Mediated Immunity and Protects Mice against a Lethal Challenge

West Nile virus (WNV) is a mosquito-borne flavivirus that is endemic in Africa, the Middle East, Europe and the United States. There is currently no antiviral treatment or human vaccine available to treat or prevent WNV infection. DNA plasmid-based vaccines represent a new approach for controlling infectious diseases. In rodents, DNA vaccines have been shown to induce B cell and cytotoxic T cel...

Assessment of humoral immune response of a Cytomegalovirus DNA-vaccine candidate in BALB/c mice

Introduction: Glycoprotein B (gB) is the major antigen for induction of humoral responses against human cytomegalovirus (HCMV) making it an attractive candidate for immune prophylaxis. In the present study, the humoral immune response of BALB/c mice to a truncated HCMV gB protein fused with GFP was evaluated. Methods: The truncated gB coding sequence was synthesized and cloned in pEGFPN1 eukary...

Comparative Evaluation of Mx and Alum as Bio and Conventional Adjuvants in Inducing Immune Responses by Influenza DNA Vaccine

Background & Objective: DNA vaccines as a new generation of vaccines require adjuvant to improve vaccine immunogenicity; adjuvants can also increase the DNA vaccine efficacy. In this study, the effects of the host’s interferon-inducible Mx protein as bio adjuvant and conventional alum adjuvant were evaluated.   Materials & Methods: The BALB/c mice were immunized by different prime-boost strate...

Protection against lethal vaccinia virus challenge by using an attenuated matrix protein mutant vesicular stomatitis virus vaccine vector expressing poxvirus antigens.

Recombinant vesicular stomatitis viruses (VSV) are excellent candidate vectors for vaccination against human diseases. The neurovirulence of VSV in animal models requires the attenuation of the virus for use in humans. Previous efforts have focused on attenuating virus replication. Studies presented here test an alternative approach for attenuation that uses a matrix (M) protein mutant (rM51R) ...

Protection of mice deficient in mature B cells from West Nile virus infection by passive and active immunization

B cell activating factor receptor (BAFFR)-/- mice have a profound reduction in mature B cells, but unlike μMT mice, they have normal numbers of newly formed, immature B cells. Using a West Nile virus (WNV) challenge model that requires antibodies (Abs) for protection, we found that unlike wild-type (WT) mice, BAFFR-/- mice were highly susceptible to WNV and succumbed to infection within 8 to 12...
